Reconstructing and Reprogramming the Tumor-Propagating Potential of Glioblastoma Stem-Like Cells
Researchers identified a core set of neurodevelopmental transcription factors (TFs) essential for glioblastoma (GBM) propagation. These TFs coordinately bind and activate tumor-propagating cell (TPC)-specific regulatory elements and are sufficient to fully reprogram differentiated GBM cells to “induced” TPCs, recapitulating the epigenetic landscape and phenotype of native TPCs. Physiologic Hypoxia Promotes Maintenance of Chronic Myeloid Leukemia Stem Cells Despite Effective BCR-ABL1 Inhibition
Given that the bone marrow hypoxic microenvironment supports hematopoietic stem cells, scientists investigated if hypoxia similarly contributes to leukemia stem cells persistence. Importantly, they found that while BCR-ABL1 kinase remained effectively inhibited by imatinib under hypoxia, apoptosis became partially suppressed. [Blood] Abstract

CD58, a Novel Surface Marker, Promotes Self-Renewal of Tumor-Initiating Cells in Colorectal Cancer
Scientists found that CD58 and CD44 were upregulated using a cDNA GeneChip, and CD44highCD58high cancer cells, the common existence of which was demonstrated by flow cytometry in multiple colon cancer cell lines and primary specimens, exhibited enhanced self-renewal ability, epithelial-mesenchymal transition ability and tumorigenicity, both in vitro and in vivo. A New Mosaic Pattern in Glioma Vascularization: Exogenous Endothelial Progenitor Cells Integrating into the Vessels Containing Tumor-Derived Endothelial Cells
Scientists found a new mosaic pattern that exogenous endothelial progenitor cells (EPCs) integrated into the vessels containing the tumor-derived endothelial cells in C6 glioma rat model. Researchers studied the effects of substrate properties including elasticity, dimensionality and matrix composition on the response of glioblastoma stem-like cells to chemotherapeutic agents. [Mol Cancer Ther] Abstract

Pancreatic Cancer Stem-Like Cells Display Aggressive Behavior Mediated via Activation of FoxQ1
Researchers found that isolated triple-marker positive cells of human pancreatic cancer MiaPaCa-2 and L3.6pl cells behave as cancer stem-like cells (CSLCs). Cobblestone-Area Forming Cells Derived from Patients with Mantle Cell Lymphoma Are Enriched for CD133+ Tumor-Initiating Cells
Researchers demonstrated that a subset of primary mantle cell lymphoma (MCL) cells co-cultured with either primary human mesenchymal stromal cells or murine MS-5 cells form in cobblestone-areas consisting of cells with a primitive immunophenotype containing the chromosomal translocation t (11;14)(q13;q32) characteristic of MCL. Design and In Vitro Evaluation of Layer by Layer siRNA Nanovectors Targeting Breast Tumor Initiating Cells
The stability, cell uptake, and release profile for small interfering RNA (siRNA) nanovectors (SNVs) were examined. In addition, SNVs reduced tumor initiating cell-related STAT3 expression levels, CD44+/CD24/EpCAM+ surface marker levels and the number of mammospheres formed compared to the standard transfection agent. miR-612 Suppresses the Stemness of Liver Cancer via Wnt/β-Catenin Signaling
Researchers revealed that microRNA-612 (miR-612) is able to suppress the stemness of hepatocellular carcinoma by reducing the number and size of tumorspheres as well as clone formation in soft agar, and to relieve drug resistance to cisplatin and 5-fluorouracil.
