EphA3 Maintains Tumorigenicity and Is a Therapeutic Target in Glioblastoma Multiforme
Researchers showed that the receptor tyrosine kinase EphA3 is frequently overexpressed in glioblastoma and, in particular, in the most aggressive mesenchymal subtype. Importantly, EphA3 is highly expressed on the tumor-initiating cell population in glioma and appears critically involved in maintaining tumor cells in a less differentiated state by modulating mitogen-activated protein kinase signaling. TGF-β Inhibition Enhances Chemotherapy Action against Triple-Negative Breast Cancer
Data suggested that chemotherapy-induced TGF-β signaling enhances tumor recurrence through IL-8-dependent expansion of cancer stem-like cells (CSCs) and that TGF-β pathway inhibitors prevent the development of drug-resistant CSCs. [J Clin Invest] Full Article

MELK-Dependent FOXM1 Phosphorylation Is Essential for Proliferation of Glioma Stem Cells
Researchers demonstrated that FOXM1 forms a protein complex with the mitotic kinase MELK in glioma stem-like cells, leading to phosphorylation and activation of FOXM1 in a MELK kinase-dependent manner. [Stem Cells] Abstract | Full Article

Cellular Plasticity Confers Migratory and Invasive Advantages to a Population of Glioblastoma-Initiating Cells that Infiltrate Peritumoral Tissue
Glioblastoma stem-like or initiating cells (GICs) have been identified in the peritumoral (PT) area, but whether these GICs have enhanced migratory and invasive capabilities compared with GICs from the tumor mass (TM) is presently unknown. Scientists isolated GICs from the infiltrated PT tissue and the TM of three patients and found that PT cells have an advantage over TM cells in 2D and 3D migration and invasion assays. An Automated High-Throughput Counting Method for Screening Circulating Tumor Cells in Peripheral Blood
This paper describes an automated high-throughput counting method for circulating tumor cells (CTCs) based on microfluidics and line-confocal microscopy. The authors also demonstrate the ability to enumerate CTCs from breast cancer patients that were positive for Her2 or CD44+/CD24, which is a putative cancer stem cell marker. [Anal Chem] Abstract

Peracetylated (-)-Epigallocatechin-3-Gallate (AcEGCG) Potently Prevents Skin Carcinogenesis by Suppressing the PKD1-Dependent Signaling Pathway in CD34+ Skin Stem Cells and Skin Tumors
Scientists investigated the chemopreventive potential of AcEGCG on 7,12-dimethylbenz[a]-anthracene-initiated and 12-O-tetradecanoylphorbol-13-acetate-promoted skin tumorigenesis in ICR mice and to elucidate the possible mechanisms involved in the inhibitory action of protein kinase D1 (PKD1) on cancer stem cells. Oxidative Stress Specifically Downregulates Survivin to Promote Breast Tumor Formation
Using in vivo models of HMLEHRAS xenografts and E2-induced breast tumors in ACI rats, researchers demonstrated that high oxidative stress downregulates survivin during initiation of tumorigenesis. Overexpression of survivin in HMLEHRAS cells led to a significant delay in tumor initiation and tumor volume in nude mice. [Br J Cancer] Abstract

Research Improving Breast Cancer Treatment by Targeting Tumor Initiating Cells
Anna Zolkiewska, associate professor of biochemistry and molecular biophysics, has received a four-year $1.245 million grant from the National Cancer Institute – at the National Institutes of Health – to study a promising breast cancer marker called ADAM12. Aduro Announces Acquisition of GVAX Assets from BioSante
Aduro BioTech, Inc. announced the acquisition of all GVAX assets from BioSante Pharmaceuticals, Inc., including intellectual property and cell lines. Aduro previously licensed two GVAX vaccines, GVAX Pancreas and GVAX Prostate, for use in combination with its Listeria-based vaccines. This new acquisition covers all uses and includes additional vaccines for multiple myeloma and breast and colon cancer and assumes rights to the existing license agreement for GVAX Melanoma.
